Plant regeneration through somatic embryogenesis and polyamine levels in cultures of grasses of Thar Desert

چکیده
Tissue culture methods for plant regeneration have been developed for Lasiurus scindicus, Sorghum halepense and Urochloa panicoides using basal portion of young inflorescence as new and a potential source(s) of explants. Highly embryogenic cultures were initiated on Murashige and Skoog’s (MS) medium supplemented with various concentrations of 2,4-dichlorophenoxyacetic acid (2-4,D). The regenerative cultures were maintained by subculture on MS medium in the dark at 26+2C. Plants were regenerated via somatic embryogenesis from the competent cell cultures on hormone-free MS basal medium or MS medium with 1mgl kinetin. Somatic seedlings were successfully transplanted to pots and grown to maturity in field. Polyamine levels were analyzed in embryogenic and non-embryogenic cultures. A correlation has been found between the individual polyamine levels and the ratio of putrescine to spermidine in the cultured tissues with plant regeneration ability via somatic embryogenesis.

Somatic Embryogenesis and Plant Regeneration in Evening Primrose (Oenothera biennis L.)
Callus induction responses and regeneration through callus-mediated SE were studied from evening primrose (Oenothera biennis L.) as an important medicinal plant of Onagraceae mainly known for its gamma-linoleic acid (GLA) content. The effects of cytokinins, 6-Benzylaminopurine (BAP) and N-(2-furfurylamino)1-H-purine-6-amine [Kinetin (Kin)] and their concentrations (0, 0.5, 1.0 and 1.5 mg/l) in ...
متن کاملSomatic Embryogenesis: An Alternative Method for in vitro Micropropagation
Plant tissue culture is an alternative method of commercial propagation and is being used widely for the commercial propagation of a large number of plant species, including many medicinal plants. Somatic embryogenesis is a process by which asexual or somatic cells are induced to form embryos in culture. Somatic embryogenesis is a multi-step regeneration process starting with formation of pro-e...

In the present study, 2, 4-dichlorophenoxyacetic acid (2, 4-D) was assessed individually for its effectiveness to induce somatic embryogenesis in tea (Camellia sinenesis L.). Embryonic axes and cotyledons explants were dissected from the seeds. Explants were cultured on Murashige and Skoog (MS) medium containing 0, 1 and 5 mM 2, 4-D alone for embryonic axes and 0, 1 and 5 mM 2, 4-D along with 0...
